Mannitol and phenytoin were used as validation standards for the comparison of culturing Caco-2 cells under manual culture conditions versus the automated Cytogration process for 21 days.

Results indicated a good agreement in mannitol and phenytoin permeability between the automated Cytogration process versus manual culture conditions.

Variation of mannitol and phenytoin permeability values obtained from Caco-2 cells under automated Cytogration processes was less than under manual conditions.
