Caco-2 Quick Screening

TESTING PARAMETERS

  • All compounds cold
  • One concentration per compound
  • One replicate per compound
  • One sample taken Apical to Basolateral side at 45 minutes
  • A summary table of the results is transferred electronically to the client

TEST CONDITIONS

  • Caco-2 cell membranes prepared with 21 to 28 days incubation in 24-well Transwell plates
  • Each set of membranes tested for membrane integrity and function by determining the permeability coefficients for mannitol and phenytoin
  • One test compound concentration
  • One sample per concentration
  • One sampling time point at 45 minutes from the receiver side
  • Sample concentration analysis will be performed by LCMS or other appropriate method
  • Methods may be modified to meet a particular test format.
